力竭运动对大鼠肾功能和肾组织紧密连接蛋白的影响
摘要:
[目的]探讨大强度力竭运动致蛋白尿负荷大鼠肾功能和肾组织紧密连接蛋白的变化及意义,为运动性肾损伤的防治及其机制研究提供新的思路。[方法]SD雄性大鼠18只,其中8只设为正常对照组,10只为力竭运动组,建立4周大强度力竭训练模型,造成大鼠持续的运动性蛋白尿负荷;检测肾功能血液指标即血清肌酐(Cre)和血尿素氮(BUN),检测肾损伤尿液指标即尿中性粒细胞明胶酶相关脂质运载蛋白(NGAL)和肾损伤分子1(KIM-1)水平变化;苏木青-伊红(HE)染色观察大鼠肾组织病理损伤情况;采用Western blot及免疫组化方法检测肾组织中紧密连接蛋白1(ZO-1)和密封蛋白Claudin-1的表达变化。[结果]4周大强度力竭运动可导致大鼠运动性蛋白尿负荷,上调肾功能标志物血清 Cre(P<0. 01)和 BUN(P<0. 01) ,尿 NGAL(P<0. 01) 和 KIM-1(P<0. 01)水平,下调ZO-1(P<0.01)、Claudin-1(P<0.05)在蛋白水平的表达。[结论]4周力竭运动可造成大鼠持续的运动性蛋白尿负荷,并导致大鼠运动性肾损伤,长时间力竭运动可能通过下调肾脏紧密连接蛋白而影响肾小管上皮紧密连接结构的完整性。
[Objective]To investigatethe changesand significanceofrenalfunction and tightjunction proteinsin ratswith high-intensity exhaustive exercise-induced proteinuria, providing new insights for the prevention and treatment of exercise-in-duced kidneyinjuryand itsmechanism study.[Methods]Eighteen maleSD ratsweredivided into anormalcontrolgroup(n=8)and an exhaustive exercise group(n=10) .A 4-week high-intensity exhaustive training model was established to induce sus-tained exercise-induced proteinuria in rats. Blood indicators of renal function, including serum creatinine(Cre)and blood ureanitrogen(BUN) , aswellasurineindicatorsofrenaldamage, such asneutrophilgelatinase-associated lipocalin(NGAL) and kid-ney injury molecule 1(KIM-1)levels, were measured. Hematoxylin-eosin(HE)staining was performed to observe pathological changes in ratkidney tissues. Western blotand immunohistochemistry methods were used to detectthe expression changes of tightjunction protein1(ZO-1)and sealing protein Claudin-1 in kidney tissues. [Results]Four weeks of high-intensity exhaus-tive exerciseinduced exercise-induced proteinuriain rats, upregulated renalfunction markersserum Cre(P<0.01) and BUN(P<0.01) , urineNGAL(P<0.01)and KIM-1(P<0.01)levels, and downregulated the expression ofZO-1(P<0.01)and Clau-din-1(P<0.05)atthe protein levelin kidneytissues. [Conclusion]Fourweeks ofexhaustive exercise can cause sustained exer-cise-induced proteinuria in rats and lead to exercise-induced kidneyinjuryin rats. Long-term exhaustive exercise may affectthe integrity ofrenaltubular epithelialtightjunction structure by down regulating renaltightjunction proteins.
