Genetic structure of weevil larva populations at geological phosphorus-rich and-deficient sites in the subtropics

亚热带地质富磷土壤生境会影响植物种群遗传结构,但是否影响植食性昆虫种群遗传结构还未见报道.在滇中高原地质性富磷土壤立地和周边典型亚热带贫磷土壤立地,选择寄生于栓皮栎(Quercus variabilis Bl.)种子的栗实象甲(Curculio davidi Fairmaire)为研究对象,进行简化基因组双酶切RAD-seq测序分析,探讨土壤磷获得性-植食性昆虫种群遗传结构的关系.结果发现:1)富磷立地象甲种群遗传多样性高于贫磷立地象甲种群;2)分子方差分析表明,6个栗实象甲种群在贫富磷立地间、种群间以及个体间分化都不显著,但是贫富磷两种立地两个象甲群体间分化显著;3)贫磷立地象甲种群与富磷立地种群间遗传距离较远,群体遗传来源组成差异较大.研究首次采用群体遗传学探讨了生长在基于地质化学的差异磷供给条件下的寄生昆虫自然群体遗传结构特征,为深入了解生物响应人为富磷化的特点、可能的生态后果以及生物进化过程提供科学数据.

Geological phosphorus-rich soil habitats in the subtropics can affect the genetic structure of plant populations,but it has not been reported whether they affect the genetic structure of herbivorous insect populations.At the geological phosphorus-rich soil sites and the typical subtropical phosphorus-deficient soil sites in the Central Yunnan Plateau,weevil larvae(Curculio davidi Fairmaire),which parasitized seeds of Quercus variabilis Bl.was selected as the research object,the double digest restriction-site associated DNA sequencing was carried out,and the relationship between soil phosphorus availability and genetic structure of herbivorous insects was explored.Results are as follows:1)genetic diversity of weevil larva population at phosphorus-rich site was higher than that at phosphorus-deficient site;2)Analysis of molecular variance showed that there was no significant genetic divergence of the 6 weevil larva populations between phosphorus-rich and-deficient sites,among populations and within populations,but there was significant genetic divergence between 2 weevil larva populations at phosphorus-rich site and phosphorus-deficient site;3)the genetic distance was far and the genetic components remained divergence distinctively between weevil larva populations at phosphorus-rich site and weevil larva populations at phosphorus-deficient site.In this study,population genetics is used for the first time to study the genetic structure of natural populations of parasitic insect growing under conditions of geochemically differential phosphorus supply,which contributes to a deeper understanding of the characteristics and possible ecological consequences of organism in response to anthropogenic phosphorus-loading,and the process of biological evolution.
