Effect of Chlorogenic Acid on LPS-induced COX-2 in Vivo and Vitro
摘要:
探讨绿原酸对脂多糖致炎症小鼠体内体外COX-2蛋白表达的影响.脂多糖滴鼻造模建立小鼠急性肺损伤模型,脂多糖诱导小鼠巨噬细胞RAW264.7建立细胞炎症反应模型,应用Weston blot法,检测小鼠肺组织内以及RAW264.7巨噬细胞中COX-2的蛋白表达.绿原酸在小鼠体内体外均能有效降低脂多糖诱导的COX-2的蛋白表达,并能促进COX-2蛋白的降解.降低脂多糖诱导的COX-2的蛋白水平是绿原酸抗炎的机制之一.
The object of this study was to explore the effect of chlorogenic acid on LPS-induced COX-2 in vivo and vitro.The mice induced by LPS was used as an acute lung injury model.The RAW264.7 cells induced by LPS was used as an cell inflammatory model.The expression of COX-2in lung tissue as well as in RAW264.7 cells was detected by Western blot.The results showed that chlorogenic acid could inhibit the expression of COX-2 protein in vivo and vitro effectively and promote the protein degradation of COX-2 protein in RAW264.7 cells.Reducing the protein level of LPS-induced COX-2 is one of the mechanisms for the anti-inflammatory action of chlorogenic acid.
